Abstract
Objective:To observation of hypoxia inducible factor-1α in hypoxic cultured osteoblasts in rats and explore its relationship with angiogenesis.Methods:The final concentration of 125umol/L cobalt chloride added into the bone cell culture medium simulated hypoxia and training at different times based group(0,3,6,9,12h) ,using reverse transcription polymerase chain reaction(RT-PCR) to detect hypoxic osteoblasts at different times in the HIF-1α and VEGF expression.Results:Osteoblasts cultured for three hours in hypoxia,the HIF-1α mRNA and VEGF mRNA expression levels increased gradually,peaked at 9 hours,then decreased,but still higher than the level before hypoxia.Conclusion:HIF-1α may regulate VEGF expression and promote angiogenesis,and this may promote the degree of hypoxia.
