Abstract
Objective To explore the apoptosis pathway of the human vascular smooth muscle cells(HVSMCs)induced by homocysteine.Methods HVSMCs were cultured in vitro.The expressions of Fas and caspase-8 protein in HVSMCs were detected by immunohistochemistry Envision two-step method and western blot.Results Immunohistochemistry of Fas protein indicate that the positive particles existed in the both membrane and cytoplasm after the treatment of Hcy.The positive particles of caspase-8 protein existed in the both cytoplasm and nucleus.The intensity of the two proteins increased in time and concentrations dependent manner(P0.05).Conclusion Homocysteine can increase the expressions of death receptors Fas in HVSMCs.Fas induce the increasing of caspase-8 protein.Death receptors-mediated signaling pathways may be one of the pathway in VSMCs apoptosis induced by homocysteine.
