Abstract
Objective To explore the effect of different doses alcohol on cardiac tissue in rats.Methods Total 140 2~3 months old SD rats were randomly divided into 7 groups: the normal control group and the different doses lavage alcohol model group: 0.8 mL/(kg·d).(Group A) 1.6 mL/(kg·d).(Group B) 2.4 mL/(kg·d).(Group C) 3.2 mL/(kg·d).(Group D) 4.0 mL/(kg·d).(Group E) 4.8 mL/(kg·d).(Group F).n=20;each group had 20 rats containing half of the male and female.Feeding methods: gavage needle was used to feed,and gavage with volume 10 mL/kg was given to control group daily.The control group was fed with 0.9% NS.The rest of the model groups were given 53° Lai Mao Alcohol.They were weighed before feeding on the very first day of the week;then weighed once a week,gavage 6 times per week and consecutive gavage for 16 weeks.Four months later,Jugular Venous blood was immediately taken from each rat for 4 mL to detect creatine kinase isozymes(CK-MB),cardiac troponin T(cTnT),High-sensitivity C-reactive protein(hs-CRP) the myocardial tissue in vivo from rat for pathological examination to observe the impact of alcohol on them.Results Compared Group A,B with control group,CK(MB,cTnT had no obvious change and it was not statistically significant(P0.05),models of Group C,D,E,F of CK-MB,cTnT levels were significantly increased,the difference was statistically significant(P0.01).Models of Group A and B compared with controls,difference of hs-CRP was statistically significant(P0.05),hs-CRP level in models of Group C,D,E,F and control group were obviously higher than model of Group A and B.The difference was statistically significant(P0.05).Models of Group D,E,F were observed in myocardial edema,myocardial cell degeneration and necrosis,and the model of Group A,B,C compared with the control group of myocardial pathological had no obvious change.Conclusion Moderate drinking can reduce hs-CRP,protecting myocardial cell;however,excessive alcohol intake [2.4 mL/(kg·d)]can lead to the rising of myocardial enzymes,increases of Caspase-3 expression,and the myocardial cell apoptosis or necrosis.
