Abstract

To provide objective basis for individualizing chemotherapy of ovarian cancer by the MTT assay as in vitro chemosensitivity test,the reformed MTT assay was adopted for chemosensitivity testing of 6 human ovarian cancer cell lines.Ten chemotherapeutic agents were used in this test.From the dose effect curve of ovarian cancer cell lines to the agents with 10 different concentrations,we obtained the values of ID 50 and AUC at ID50 to evaluate the potency.Results showed that ①Chemosensitivity had no positive relationship with the pathological classifications and primary focus sites.But,for different cell lines,the difference of chemosensitivity was significant ( P 0.001);②Using the assay ID 50 ,5 cell lines were sensitive to IFM,4 cell lines were sensitive to CPM,ACT D and 2 sensitive to P MMC,VBL.Using the assay AUC,6 cell lines were sensitive to IFM,4 sensitive to NK611 and 3 sensitive to CPM.The results suggest that the reformed MTT assay can be used as a in vitro chemosensitivity test which has great significance of directing clinical chemotherapy for patients with ovarian cancer.
