Influence of Losartan on Transforming Growth Factor-β1 and Connective Tissue Growth Factor in Rats with Diabetic Nephropathy

Abstract

Objective To investigate expression of transforming growth factor-β1(TGF-β1)and connective tissue growth factor(CTGF) protein in renal tissues,and detect the levels of urinary TGF-β1 and CTGF in rats with diabetic nephropathy(DN).To observe the influence of losartan on expression of the two protein in renal tissue and excretion in urine.Methods Wistar rats were treated by intravenous injection of streptozotocin after right nephrectomy to induce DN rat model.The DN rats were randomly divided into two groups:DN experimental group and losartan treated group.The expression of TGF-β1 and CTGF in renal tissue were determined with immunohistochemical staining,urinary TGF-β1 and CTGF were assayed by enzyme-linked immunosorbent assay(ELISA) at 6,12 weeks respectively.Results Compared with losartan treated group,urinary protein excretion and the protein expression of TGF-β1 and CTGF significantly increased(P0.01) and the relative area of interstitial fibrosis significantly enlarged in the experimental group(all P0.01),the relative area of glomerular mesangium and basement membrane protein enlarged(P0.05)at 12 weeks.Compared with at 6 weeks,the levels in urine and the protein expression in renal tissue of TGF-β1 and CTGF,the relative area of interstitial fibrosis significantly increased in experimental group at 12 weeks(P0.01).The expression of TGF-β1 protein in glomerular and renal tubular increased in losartan treated group at 12 weeks(P0.05 or P0.01 respectively).A significant positive correlation was observed between the protein expression of TGF-β1 and CTGF in renal tissue(r=0.85 P0.01),and the excretion of TGF-β1 and CTGF in urine also(r=0.76 P0.01).Conclusions The expression and urinary protein excretion of TGF-β1 and CTGF protein significantly increase in DN rats.Losartan can down-regulation the protein expression of TGF-β1 and CTGF in renal tissue,decrease the urinary excretion to protect kidney.
