Study on expression and clinicopathologic characteristics of miR-195 and miR-497 in colorectal cancers

Abstract

Objective MicroRNA(miRNA) participate in various physiological and pathological events as post-transcriptional regulators.Studies have shown that the disorders of miRNAs on the regulation of tumor suppressor/oncogene,promote the development and progression of tumors.The aim of this study is to elucidate the miR-195 and miR-497 expressions in colorectal cancers and analyze their correlation with clinicopathologic features.Methods Real time PCR was used to quantify the miR-195 and miR-497 expressions in 57 pairs of colorectal cancer and normal mucosa samples,and the relationship of their expressions and clinicopathologic features was analyzed.Results The miR-195 and miR-497 expressions in colorectal cancers were down-regulated compared to normal mucosa(P0.01,respectively).Furthermore,the reduced expression of miR-195 occurred more often in patients with lymph node metastasis and advanced tumor stage.Kaplan-Meier survival analysis indicated that patients with reduced miR-195 had a poor overall survival(P0.01).There was no relationship between miR-497 and gender,TNM stages,local invasion,lymph metastasis,distant metastasis or cell differentiation(P0.05,respectively).ConclusionBoth miR-195 and miR-497 were down-regulated in colorectal cancers.Reduced expression of miR-195 may play a certain role in the development of colorectal cancer.MiR-195 may be a potential indicator in the auxiliary diagnosis and prognosis of colorectal cancers.
