Abstract

Objective: To establish Parkinson's rat model mimicking different stages in the evolution of Parkinson's disease . Methods: In three different lesion groups 6-OHDA was injected into substantia nigra, lateral substantia nigra pars compacta and striatum. The behavioural and morphological change were observed after the microinjection with different amount of 6-OHDA solution. Results: ⑴Unilateral subtantia nigra lesion in rat causes dopaminergic neurons loss more than 95%.⑵Unilateral lateral substantia nigra lesion in rat causes dopaminergic neurons loss more than 70%. ⑶Unilateral striatum lesion in rat causes dopaminergic neurons loss less than 50%. Conclusion: The different site and amount of microinjection 6-OHDA can establish PD rat mimicking different symptomatic stage in PD. It may be particularly useful as an experimental model in studies of experimental interventions aimed at neuroprotection, regeneration or functional recovery in the nigrostriatal DA system.
