Abstract
Objective To develop a method to prepare high quality genomic DNA of A.officinarum Hance for downstream molecubiological research.MethodsFour protocols based on modified CTAB method was performed to extract genomic DNA from A.officinarum Hance.ResultsThe OD260/OD280 ratios of genomic DNA extracted with the fourth protocol of modified CTAB method ranged from 1.8 to 2.0,the yields of DNA sample were in the range of 0.54~1.049 μg/mg dried leaf tissue.DNA samples were completely digested with restriction enzymes(EcoRⅠ and Mse Ⅰ) and were successfully used for AFLP.ConclusionThe DNA samples extracted with the fourth protocol of modified CTAB method are with high quality and suitable for molecubiological research.
