Abstract
Objective:To investigate the effect of ERβ on the expression of OPG in human periodontal ligament (HPLF)cells. Method:We employed a short interfering RNA (siRNA) technique to inhibit ERβ expression in HPLF cells and cells were cultured with saturating concentration of 17β-estradiol (10-7 mol/L) for 48 hours. Changes of the expression of OPG were determined by RT-PCR and Western blot analysis. Result:It was shown that estradiol caused an increase in OPG expression in HPLF cells. However,estradiol had no effect on the expression of OPG in HPLFsiERβ cells. Conclusion:These results indicate that ERβ is of major importance for estrogen-mediated effects on gene transcription in HPLF cells and estrogen influences the expression of OPG in HPLF cells via ERβ.
