Expression of gene BRCA1 and BRCA2 in sporadic invasive ductal carcinoma and its correlation with clinical medicine

Abstract
Objective:To investigate the level of breast cancer susceptibility gene 1(BRCA1) and BRCA2 in sporadic invasive ductal carcinoma and to analyze its relationship with other clinicopathologic parameters.Methods:RT-PCR was used to detect the level of gene BRCA1 and BRCA2 in 95 cases of sporadic invasive ductal carcinoma tissue samples and 28 adenosis tissue samples.The clinicopathologic parameters including tumor size,axillary nodal status,estrogen receptor and HER-2 expression were obtained from pathological documents.The level of BRCA1/BRCA2 was compared between the sporadic invasive ductal carcinoma tissues and adenosis tissues.In sporadic invasive ductal carcinoma,the correlation between BRCA1/BRCA2 and other clinicopathologic parameters mentioned above was further analyzed.Results:The positive rates of BRCA1 gene were 50.5%(48/95)in breast cancer tissues and 89.3%(25/28)in adenosis tissues,while in breast cancer adjacent tissues,the rates were 66.3%(63/95).The positive rates of BRCA2 gene were 52.6%(50/95)in breast cancer tissues and 92.9%(26/28)in adenosis tissues,while in breast cancer adjacent tissues,the rates were 72.6%(69/95).The positive rates of BRCA1 and BRCA2 were lower in breast cancer tissues than in cancer adjacent tissues and adenosis tissues,with a significant difference(P0.05).The gene BRCA1/BRCA2 was negatively correlated with lymph node metastasis,and negatively correlated with the expression of estrogen receptor and HER-2 expression(P0.05).No correlation was found between BRCA1/BRCA2 and tumor size(P0.05).Conclusion:The gene BRCA1/BRCA2 in breast epithelium may play an important role in the development of breast carcinoma.To detect them helps to choose patients at high risk of developing breast carcinoma and achieve early diagnosis.The gene BRCA1/BRCA2 may be a potential marker for estimating the biological behavior and prognosis of breast cancer.
