Synthesis of Artificial Antigens for Sulfanilamides and the Preparation of Its Antiserum

Abstract

To lay a foundation for the preparation of sulfanilamides' monoclonal antibody and the development of its gold-immunochromatographic assay,Hapten H was synthesized and its antiserum were prepared.Nacetylsulianily chloride and para-amino benzoic acid were used to synthesize Hapten(H),and it was coupled to carrier protein bovine serum albumin(BSA) and ovalbumin(OVA) by using the mixed acid ahhdride method.The conjugates were identified by UV absorption method to calculate the coupling ratio and the protein concentration.H-BSA was used as immunogen to inject the rabbits to prepare antiserum.The affinity and the titer of the antiserum were detected by using the H-OVA as coating antigen.Sulfanilamides' artificial antigens were synthesized successfully.The protein concentration of H-BSA was 8.46 mg/mL or 10.53 mg/mL for H-OVA,respectividy.The coupling ratio of H to BSA was 13∶1 and to OVA 9∶1.The titer of antiserum was 1∶256 000.
