Abstract

Objective:To study the influence of arsenic trioxide (As_2O_3) injection combined with/or fiuorouracil (5-FU) on the growth and telomerase activity of human large intestine carcinoma cell lines.Methods:The influence of growth on human large intestine carcinoma cell lines LoVo and CoLo-320 induced by As_2O_3 injection combined with/or 5-FU was analysed by 3-(4,5-dimethytthiazole-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT) assay. Telomerase activity of the carcinoma cells was measured by the method of Repeat Amplitcation Protocol (TRAP) combining with Polyacrylamide Gel Electrophosis (PAGE) Silver Staining method before and after treatment of As_2O_3 injection combined with/or 5-FU.Results:As_2O_3 injection combined with 5-FU may obviously improve the anti-tumor effeet on human large intestine carcinoma, and may strengthen inhibiting the tolemerase activity comparing each individual drug.Conclusion:As_2O_3 injection combined with 5-FU may obviously improve the anti-tumor effect on human large intestine carcinoma, and whose mechanism maybe related to its strengthen inhibiting the tolemerase activity.
