Determination and clinical significance of adenine nucleotide translocator 3 gene expressionon peripheral blood mononuclear cells from patients with primary biliary cirrhosis

Abstract
[Objective] To explore the correlation of adenine nucleotide translocator 3(ANT3) gene expression level with primary biliary cirrhosis(PBC).[Methods] Based on fluorescent Taqman-MGB technology,real-time fluorescent semi-quantification reverse transcriptional PCR was established,according to the difference of threshold cycles(ΔCT) of ANT3.ANT3 mRNA of PBMCs in 30 healthy people(control group) and 30 PBC patients(observed group) were measured.Meanwhile,the concentrations of γ-glutamyltransferase(γ-GT) and alkaline phosphatase(ALP) were determined.[Results] ANT3 mRNA level was significantly higher in observed group than in control group (ΔCT =9.4±0.8,P0.01),which indicated the concentration of the original template of ANT3 gene in observed group was 2.3 folds as much as that of control group.The concentrations of γ-GT and ALP were elevated in observed group,which were associated with gene expression of ANT3(r=-0.92 and r=-0.86,P0.05).[Conclusions] ANT3 gene expression level of PBMCs in PBC patients was significantly elevated and negatively associated with concentrations of γ-GT,ALP,which indicated determination of ANT3 gene played a pivotal role in pathogenesis,diagnosis and course of PBC.
