Abstract
Objective To study the influence of defibrol on the levels of platelet activating factor(PAF)and the expression of PAF receptor gene after focal ischemic reperfusion in rats. Methods The unilateral middle cerebral arteries of rats were occluded for 6 h by an intraluminal filament and were reperfused at 6 h postoperatively. Reverse transcription polymerase chain reaction(RT-PCR)was applied to evaluate the expressions of PAF receptor gene in the ischemic penumbra tissue respectively. Meanwhile, Elisa was applied to evaluate the levels of the corresponding PAF in blood plasma. Results The level of PAF increased obviously in the reperfusion group and ischemic group of the normal saline (NS) groups, but the expression of PAF receptor gene decreased. There were obvious statistical difference between NS groups and control groups. The level of PAF decreased in the defibrol groups, and the expression of the PAF receptor gene increased. There were conspicuous statistical difference between NS groups and defibrol groups. Conclusion Defibrol can reduce the level of PAF in plasma after cerebral ischemic reperfusion, Defibrol may reduce the expression of PAF receptor gene in the ischemic penumbra tissue after cerebral ischemic reperfusion.
