Effect of proanthocyanidins on the expression of the NF-κB/p65 protein and some proinflammatory factors in paws and serums of CIA rats

Abstract

Objective To investigate the inhibitory effect of proanthocyanidins on the expression of NF-κB/p65 protein in the tissue of Type Ⅱ collagen-induced arthritis(CIA) rat′s paws,and the effects of proanthocyanidins on the contents of TNF-α、IL-1β in CIA rat′s serums.Methods CIA model in SD rats was established.The proteins expression of NF-κB/p65 and MMP-9 in the skin of the rats paws was analyzed with immunohistochemistry.The contents of TNF-αand IL-1β in CIA rats serum were measured with ELISA.Results Proanthocyanidins can down-regulate the expression of NF-κB/ p65 protein and decrease level of TNF-αand IL-1β in CIA rats serums.Conclusion The anti-inflammatory mechanisms of proanthocyanidins may be related to decreasing the protein expression of NF-κB/p65 and suppressing proinflammatory factors,i.e.,TNF-α and IL-1β in CIA rats.
