Abstract
OBJECTIVE To investigate the regulating effects of Sinitang on nitric oxide(NO)and endothelin(ET)levels in blood and the expressions of endothelin receptor-A(ETRA),nitric oxide synthesis(NOS)in main target-organs in renal hypertensive rats.METHODSA total of 24 healthy female Wistar rats were randomly divided into normal control(NC)group 8 cases,renal hypertension(RH)group 8 cases and Sinitang treatment(ST)group 8 cases.The renal artery stenosis hypertensive models were established according to double-clip renal artery.The ST group is given Sinitang through intragastric administration,while the RH group is given isotonic sodium chloride at the same time.The blood pressure of rats was measured by RBF-2 tail blood pressure meter.The content of nitric oxide(NO)in serum was determined by nitrate reductase assay;and the contents of endothelin(ET)in plasma were determined by radioimmunoassay.The expressions of endothelin receptor-A(ETRA),nitric oxide synthesis(NOS)in heart,brain and kidney tissue of each group were determined by immunochemistry assay.RESULTS①In ST group,the blood pressure was lower than that in NC group(P0.05).②The NO in serum and the ET in plasma were higher than those in NC group(P0.05).The blood pressure of rats was reversibly to NO serum level.③The expression levels of ETRA in target organs in RH group were higher than those in NC group(P0.05),while its level in ST group was lower than that in RH group in kidney and heart(P0.05),but no significant difference comparing to RH group in brain(P0.05).④The expression levels of iNOS in kidney and brain in RH group were higher than those in NC group,while its level in ST group was lower than that in RH group.The expression level of eNOS in heart in RH group was lower than that in NC group,while its level in ST group was higher than that in RH group.CONCLUSIONSinitang might play a regulating effect on blood pressure in hypertensive rats by adjusting the expression levels of vasoactive substances in main target-organs and the vasoactive substance levels in blood.
