Effects of Adenosine Preconditioning on Heme Oxygenase-1 Expression in Myocardial Ischemia/Reperfusion Injury in Rats

Abstract
Objective: To investigate the role of heme oxygenase-1(HO-1) in adenosine preconditioning against myocardial ischemia/reperfusion injury in rats.Methods: Eighteen male SD rats were randomly divided into three groups(n=6): control group(group C),ischemia/reperfusion group(group IR),and adenosine preconditioning+ischemia/reperfusion group(group AP).The left anterior descending coronary artery was ligated for 30 min and loosed for 120 min to establish the model of myocardial ischemia/reperfusion injury.At the end of 120 min reperfusion,the expression of HO-1 in myocardium was measured by immunohistochemistry.The infarct area,MDA content and SOD activity in myocardium and serum TNF-α concentration were determined.Results: Compared with those in group C,the infarct area,myocardial MDA content,and serum TNF-α concentration were increased,SOD activity was decreased(P0.05),but HO-1 expression had no changes(P0.05) in group IR.Compared with those in group IR,the infarct area,myocardial MDA content and serum TNF-α concentration were decreased(P0.05),along with increased SOD activity and HO-1 expression(P0.05) in group AP.Conclusion: Adenosine preconditioning protects myocardium against ischemia/reperfusion injury,and up-regulation of HO-1 expression may be involved in this protecting process.
