Effects of Proliferation Inhibition and Apoptosis Induction of Curcumol on SW1116 Cells and Its Mechanism

Abstract

To investigate the inhibitory effects of curcumol on proliferation inhibition and apoptosis induction in human colon carcinoma SW1116 cells and its possible mechanism,the proliferation inhibition was measured by MTT assay.The flow cytometer was used to analyze the apoptosis rate.Colorimetric method was employed to detect the activity of caspase-3.The expression of Bc l—2 and Bc l—XL proteins were measured by Western blotting assay,SW1116 cells growth and induced their apoptosis in dose and concentration-dependent manners are curcumol inhibited.After treatment with curcumol for 48 hours or 72 hours,the activity levels of caspase-3 significant increased.Furthermore,the expression levels of Bc l—2 and Bc l—XL could be decreased by curcumol in SW1116 cells.It is conclused that curcumol can inhibit the growth and induce the apoptosis of SW1116 cells significantly,which may be related with regulating the expression levels of Caspase-3,Bc l—2 and Bc l—XL.
