Abstract
To analyze the genetic diversity of the genomic DNA in Oncomelania hupensis distributed in different endemic areas for schistosomiasis.The genomic DNA extracted from the adults of O.hupensis distributed in Ningguo and Wuhu region were amplified by Restricion Fragment-RAPD(RF-RAPD).Moreover,the gel-purified RF-RAPD bands were isolated and sequenced.Sequence alignment and the motifis of the RAPD bands with similar mobility were analyzed with DNAsis.The genomic DNA of O.hupensis from Ningguo and Wuhu region were successfully amplified by RF-RAPD.Sequence alignment of four pairs of RF-RAPD bands with similar mobility between different O.hupensis showed that each pair of RF-RAPD bands were differed in GC content,and their similarities were in the arrange from 48% to 53%.It is demonstrated that there are polymorphism in the length and construction of RF-RAPD bands which has similar mobility.In addition,there are significant differences in the type and number of transcription factors in the RF-RAPD bands in O.hupensis distribute in Ningguo region as well as Wuhu region.Though there are similarity in their finger printing of the genomic DNA,gene variance is distinguished between the RF-RAPD bands which have similar mobility in O.hupensis from different endemic areas for schistosomiasis.It should be further investigated whether these genetic variation is correlation with the susceptibility of O.hupensis to miracidium of Schistosoma japonicum.These findings are useful for the further studies on the interactions between intermediate snail host and S.japonicum.
