Celecoxib-induced anti-proliferation and apoptosis of human lung adenocarcinoma cell line A549

Abstract

Objective To investigate the effect of celecoxib,a selective COX-2 inhibitor,on human lung adenocarcinoma cell line A549,and to explore its molecular mechanism.Methods After incubating K562 cells with increasing dose of celecoxib in vitro,the cell viability was analyzed with MTT assay,apoptosis of cells was detected by TUNEL,and caspase-3 activity was detected by caspase-3 activity detection kit.Results Celecoxib effectively inhibited the proliferation of A549 cells in a concentration-dependent way,and the IC50 of A549 cells was 33.98 μmol·L-1.TUNEL assay indicated that the rate of apoptosis was significantly different in cells with or without celecoxib(P0.05).The cellular caspase-3 A405 of A549 cells treated with celecoxib was obviously higher than that of the control group(P0.05).Conclusion Celecoxib can inhibit the proliferation and induce the apoptosis of A549 cells in a concentration-dependent manner.The regulation of apoptosis induction is related to the caspase-3-mediated pathway.
