Abstract
Objective To investigate the impact of celecoxib,a selective cyclooxygenase-2(COX-2)inhibitor,on growth and apoptosis of multiple myeloma(MM) cell line U266 and explore the molecule mechanism of celecoxib inducing U266 cells apoptosis through comparing apoptosis rate and expressions of caspase-3 of U266 cells stimulated by celecoxib alone and celecoxib adding caspase-3 inhibitor(Ac-DEVD-FMK).Methods After MM cell line U266 was treated with celecoxib of different concentrations,CCK-8 extrinsic drug screening method was used to detect whether celecoxib had inhibition effect on proliferation of U266 cells in vitro and it's effect intention.Flow cytometry staining with Annexin V-FITC was used to detect the U266 cells apoptosis.Western blot was used to detect the caspase-3 expression in different samples.Results Celecoxib could inhibit the growth of U266 cells when its' concentration was more than 40 μmol/L with a dose-dependent manner,and median inhibit concentration(IC50) was 66.83 μmol/L.Celecoxib could induce apoptosis of U266 cells when its' concentration ranged from 40 μmol/L to 120 μmol/L with a dose-dependent manner.The apoptosis of U266 cells treated with Celecoxib could be inhibited significantly when the activation of Caspase-3 was inhibited by Ac-DEVD-FMK.Conclusions Celecoxib can induce the MM cell line U266 apoptosis,and its mechanism might associated with the caspase-3-depended signal passway of apoptosis regulation.
