Abstract
Objective: To investigate the expression of nuclear factor-kappa B(NF-κB) in the kidney of diabetic rats induced by streptozocin(STZ),and to study the effects of pyrrolidine dithiocarbamate(PDTC) on the expression of nuclear factor-kappa B(NF-κB) in the renal tissue of diabetic rats.Methods: Sixty male Wistar rats were divided randomly into three groups: normal control group(Group C,n=20),diabetic model group(Group M,n=20) and diabetic rats with treatment of PDTC(a inhibitor of NF-κB)(Group P,n=20).The rats were killed at the end of 4 and 8 weeks later respectively.All rats were weighed and 24 h urine was collected for determination of urine microprotein and urine albumin excretion rate(UAER).Blood samples were collected from heart of all rats for blood glucose,glycosylated hemoglobin,creatinine(Cr),BUN,total cholesterol,triglyceride and lipoprotein measurement.Kidneys were isolated and weighed,and the expression of NF-κB were detected by immunohistochemistry test.Results: The expression of NF-κB in STZ induced diabetic rats was significantly higher than in Group C(P0.01),and the treatment with PDTC significantly decreased the enhanced expression of these two factors(P vs.M,P0.01),although the expression of NF-κB was still higher in Group P than that in Group C(P0.01).The expression of NF-κB in kidney of diabetic rats was positively correlated with the level of UAER.Conclusion: Down-regulation of the expression of NF-κB could decrease the level of UAER.The further research on NF-κB will be helpful to prevent the development of diabetic nephropathy.
