Abstract
Objective: To observe the protective effect of Propofol on lung ischemia-reperfusion injury in rats.Methods: Thirty male SPF Sprague-Dawley rats were randomly divided into 3 groups: sham group(n=10,the left chest was opened without clamping the left hilum of lung),lung ischemia reperfusion injury group(n=10,the injury was induced by clamping the left lung hilum for 45 min followed by 120 min reperfusion),Propofol group[n=10,Propofol(5 mg·kg-1)was injected though femoral vein 30 min before clamping left lung hilum,then Propofol(50 mg·kg-1·h-1)was injected with the trace pump.Left lung hilum was clamped for 45 min and followed by 120 min reperfusion].Three groups were given tracheal intubation,mechanical ventilation before opening chest.At the end of the reperfusion,the blood sample was withdrawn from the left ventricals to test the PaO2,and the left lung was harvest.The injury of issue and cell were observed by light microscope and the pulmonary edema was evaluated through lung wet-to-dray(W/D) ratio.The content of malondialdehyde(MDA),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6) and superoxide dismutase(SOD) activity in lung tissues were measured by ELISA.Results: Compared with sham group,the W/D ratio,MDA、TNF-α、IL-6 content in lung ischemia reperfusion injury group and Propofol group were significant higher(all P0.05),while the PaO2 and SOD activity were reduced(all P0.05).As compared with lung ischemia reperfusion injury group,the W/D ratio,the content of MDA,TNF-α,IL-6 in Propofol group were decreased,while the PaO2 and SOD activity in Propofol group were increased notably(P0.05).Histopathology showed that lung tissue was slightly damaged in sham group,baldly damaged in lung ischemia reperfusion injury group and the Propofol group was lighter than that in lung ischemia reperfusion injury group.Conclusions: Propofol can prevent SD rats lung ischemia reperfusion injury to some extent by removing oxygen free radicals and inhabiting inflammatory factor.
