The Protective Effect of Ulinastatin on the Lung-subjected to Ischemia-Reperfusion Injury in Rats

Abstract

Objective To probe into the protective effect of ulinastatin (UTI) on the lung subjected to ischemia-reperfusion injury in rats. Methods A model of ischemia-reperfusion injury of the study .The animals were randomly divided into 3 equal groups: Group Ⅰ(control, rats of this group received no treatment), Group Ⅱ(rats of this group were given each an IV injection of 6 kU·kg -1 of UIT 30 min before ischemia), Group Ⅲ(rats of this group were given each an IV injection of 12 kU·kg -1 UTI 30 min before ischemia). Blood samples were taken from the left and right atria before clamping and 30 min after loosening of the left hilus of the lung for white cell counting. Sample of lung tissue were taken before ischemia and 2 h after reperfusion for the calculation of the wet/dry weight ratio and determination of the content of malonyldialdehyde (MDA). The lung tissue was also subjected to histopalhological examination. Results The ratio of the right/left atrium white cell count and the wet/dry weight ratio of the lung after reperfusion were significantly lower in rats of group Ⅱ and group Ⅲ treated low and high doses of UTI,respectively,as compared with those of rats of group Ⅰ not treated with UTI(P0.05).The content of MDA in the lung tissue after reperfusion in rats of group Ⅲ treated with high dose of UTIwas significantly lower than that in rats of group Ⅰuntreated with UTI(P0.05).The pathological changes were not so serious in rats of group Ⅱ and group Ⅲ as compared with those in rats of group Ⅰ. Conclusion Ulinastatin was shown to exert significant protective effects on the murine lung subjected to ischemia-reperfusion injury.
