Rapid DNA Fingerprinting of Rice Germplasm Resources by SRILS Uniprimers

Abstract

In this study,the genetic variability and relationships among 265 rice germplasm accessions from the Genebank of Philippine Rice Research Institute were analyzed through DNA fingerprints generated by using two Uniprimers: Seoulin Research Institute Life Science-6(SRILS6) and Seoulin Research Institute Life Science-8(SRILS8).The DNA fingerprints showed that two SRILS primers generated 55 polymorphic bands in the germplasm samples with an average of 33.3 bands per accession,and the average percentage of polymorphic bands generated by the two Uniprimer markers was 60.5%.Three dendrograms were generated based on banding profiles generated by SRILS primers and their combination,and they divided the samples into two nearly even major groups: Group I and Group II.The dendrogram analysis showed that the two SRILS primers could distinguish all 265 accessions,and revealed a high level of genetic diversity among the accessions.The clustering patterns and the compositions of the groups were remarkably similar among the three dendrograms,their matching degree being 91%~95%.This indicated that the genomic regions amplified by SRILS markers might have undergone mutations at comparative rate so as to manifest highly congruent genetic trees.The studied SRILS marker system could be ideal tool for rapid DNA fingerprinting and diversity analysis of rice germplasm resources,and hence,it could be an important adjunct for the management of rice germplasm resources such as germplasm identification,diversity analysis and assembly of core collections.
