Abstract
A new method was established to determine the entrapment efficiency of a nanostructured lipid carrier(NLC) loading paclitaxel. An RP-HPLC was developed. The paclitaxel and its preparations were separated on C18 column using a mobile phase of water-acetonitrile gradient. The UV-detector was at 227 nm. The fluorenscence detector was used with excitation wavelength of 428 nm and emission wavelength of 515 nm.The linear range for the concentration of paclitaxel was from 3.0 mg/L to 80.0 mg/L,and the regression equation was Y=42100ρ+8250,r2=0.9999.The entrapment efficiency of the preparation was 68.78%.The method could be used to quantitate paclitaxel loaded in the NLC and determine the entrapment efficiency of the preparations.
