Abstract

Objective:To study the effect of berberine hydrochloride,at different degree,on Hela P10s-11F metastasis and explore the mechanism.Methods:Different concentrations of berberine hydrochloride were added to human cervical carcinoma Hela cells,and inhibition of cell proliferation was detected by MTT colorimetric method;Transwell chamber was applied for Artificial reconstituted basement membrane invasion and motion experiment,and the effect of berberine hydrochloride on the invasion and metastasis of Hela cells influence was observed;Immunohistochemistry SABC was used to investigate the expression of MMP-9 and NF-κ Bp65 in Hela cells.Results:MTT assay showed that berberine hydrochloride can inhibit the cervical cancer Hela cells in vitro(P0.05),and in a dose,time-dependent(P0.05),at the same time,invasion and migration of cells decreased significantly;immunohistochemistry SABC method showed that berberine hydrochloride can lead to downregulation of MMP-9,NF-κ Bp65,and compared with the control group,the difference was statistically significant(P0.01).Conclusion:Berberine hydrochloride has obvious inhibitory effect on the proliferation of cervical cancer Hela cells in vitro;berberine hydrochloride could decreased the Hela of human cervical carcinoma cell invasion and metastasis,and its mechanism may be related to the decrease of MMP-9,NF-κ Bp65 expression.
