Abstract
The explants used for transformation were stem segments of potato.The factors affecting the transformation frequency(concentration of antibiotics and Agrobacterium,co-culture time,etc.) were studied with Agrobacterium tumefaciens.The results indicated that there was better screening to callus and buds when concentration of kanamycin was 100 mg·L-1.When concentration of Agrobacterium cells was 0.5 OD600,3-day co-culture led to an increase in transformation efficiency.But during rooting stage kanamycin concentration should be 50 mg·L-1.Carbenicillin could better inhibit growth of agrobacterium.Results of PCR proved that the objective gene had been integrated into potato genome.
