Comparison of RBL-2H3 Cell and Primary Mast Cell Degranulation Induced by Xuesaitong Injection

Abstract

Objective To investigate the similarities and differences of degranulation in anaphylactoid reactions induced by Xuesaitong Injection(XI)between RBL-2H3 cells and rat peritoneal primary mast cells,and to evaluate the significance of RBL-2H3 cells and rat peritoneal primary mast cells in anaphylactoid test.Methods RBL-2H3 as well as rat peritoneal mast cells were incubated with XI,Compound 48/80 and Tween-80 for one hour.And then we measured β-hexosaminidase release rate,histamine release rate and their release content for the analysis of the differences of degranulation between RBL-2H3 cells and rat peritoneal primary mast cells.We also investigated the effect of different concentrations of XI on degranulation.Results The two kinds of cells showed degranulation after direct stimulation of XI,Compound 48 / 80 and Tween 80.And the degree of degranulation in the two kinds of cells was dose-dependent after treatment with different concentrations of XI.Primary mast cells presented higher β-hexosaminidase release rate,histamine release rate and histamine release content than RBL-2H3 cells,and the differences were significant(P 0.01).Conclusion RBL-2H3 cells and mast cells have the same trend of degranulation in the anaphylactoid reaction.Both of them can be used to evaluate the anaphylactoid reaction induced by traditional Chinese medicine injection,and RBL-2H3 cells are of convenient application while primary mast cells have higher sensitivity.
