Abstract
[Objective] To observe the inhibition of RNA interference(RNAi) targeting the HBV core gene on HBV replication.[Methods] Nt2021-2049 of HBV genome was selected as the target sequence,the respective sense and antisense oligonucleotide were synthesized,annealed and was cloned into the shRNA expressing plasmid,which was then cotransfected the HepG2 cell with the HBV plasmid,afterwards,whose inhibition on HBV was observed. [Results] Replication of HBV and expression of HBeAg were inhibited significantly.[Conclusions]The chosen target region is effective in inhibiting HBV replication by RNAi.
