Abstract
Objective To investigate the effects of sodium dimercaptopropansulfonate(DMPS) on esophageal cancer cell line EC109 in vitro and on mice bearing Ehrlich ascites cancer.Methods The esophageal cancer cell line EC109 cultured in vitro was applied to test the effects of DMPS with different concentrations on the growth and proliferation of the cancer cells by means of MTT method,and also to test the apoptosis induction of DMPS with different concentrations via fluorescence microscope and with cell apoptosisHoechst33258 kit;the effects of DMPS with different concentrations on the survival time of mice bearing Ehrlich ascites cancer was investigated.Results In vitro,DMPS,at the final concentrations of 8,4,2 and 1mmol/L,significantly stimulated EC109 cells growth and proliferation when administered 24h(P0.01) and 48h(P0.01) later,respectively;while 72h after administration,all concentrations of DMPS significantly inhibited the growth and proliferation of EC109 cell(P0.01).DMPS,at the final concentration of 8,4,2 and 1mmol/L,had no significant effects on the number of apoptotic EC109 cells when administered 24h and 48h later,respectively(P0.05).However,all concentrations of DMPS significantly increased the number of apoptotic EC109 cells when administered 72h later(P0.01).In vivo,DMPS administered daily intraperitoneally at a dose of 75mg/kg body weight and 150mg/kg body weight for 10 days,significantly lengthened the survival time of mice bearing Ehrlich ascites cancer(P0.01).Conclusion DMPS significantly suppresses the growth and proliferation of esophageal cancer cell line EC109 in vitro 72h after administration,possibly due to induction of apoptosis,and DMPS daily administered intraperitonealy for 10 days could lengthen the survival time of mice bearing Ehrlich ascites cancer.
