Effect of Nano-Fe_3O_4 on Hematological System in Mice

Abstract

Objective To observe the effect of nano-Fe3O4 on hematological system in mice.Methods Seventy-two adult male ICR mice were randomly divided into four groups:control group(high pure water),low(10 mg/kg,1/16 LD50),moderate(20 mg/kg,1/8 LD50),high(80 mg/kg,1/2 LD50) dose group of nano-Fe3O4,6 mice in each group.Nano-Fe3O4 was given through tail vein injection,the volume of injection was 10 ml/kg,the injection rate was about 0.02 ml/s.The contents of WBC,RBC,HGB and PLT change in blood were determined on 1,3,7 day after injection.Results No significant difference was seen in the count of WBC between the treatment groups and control group after 1 day;for moderate and high dose group,it was significantly lower compared with control group and low dose group after 3 day(P0.05);and the high dose group was lower significantly than the moderate dose group(P0.05);after 7 days,the treatment groups were significantly lower than that in control group(P0.05).The counts of RBC in the treatment groups were significantly lower than that in control group after one day(P0.05);after 3 days,they increased gradually;after 7 days,in high and moderate dose group they were significantly higher those in low dose group and control group(P0.05).The change of HGB was similar with the change of RBC after injected nano-Fe3O4.The content of PLT in high dose group were significantly lower than that in control group after one day;after 3 days,in the treatment groups they were significantly lower than that in control group(P0.05);after 7 days,the high dose group it was significantly lower compared with the other groups(P0.05).Conclusion In the experimental condition,nano-Fe304 may have an adverse effect on mice hemogram.
