The influence of different oxygen supply conditions to the metabolic flux of L-glutamic acid biosynthesis in Corynebacterium glutamicum S9114

Abstract

The metabolic flux balance model of biosynthesis of L-glutamic acid by Corynebacterium glutamicum S9114 was constructed.Metabolic flux distribution was calculated with combination of the model and situation of extracellular metabolites of S9114 in fermentation phase of medium to later under capacity of dissolved oxygen by 10% and 30%.And ideal glutamic acid metabolic flux distribution was obtained with linear program of MATLAB software.The results of metabolic flux analysis indicated that synthesis of glutamic acid will be increased when increase metabolic flux of CO2 anaplcrotic reactions and decrease metabolic flux of TCA cycle and HMP pathway.Dissolve of oxygen was the key factor of fermentation control,and increase of rate of dissolved oxygen in medium in later phase of fermentation would improve glutamate biosynthesis and inhibit production of by-product amino acids.Main by-product lactic acid occupied certain ratio in ideal glutamic acid metabolic flux distribution,which indicated that lactic acid might play a role in the process of glutamate biosynthesis.
