ULTRASONIC EVALUATION OF PERINDOPRIL AND SPIRONOLACTONE ON CARDIOVASCULAR REMODELING AND FUNCTION IN HYPERTENSIVE RATS

Abstract
Objective:To investigate the structural and functional changes of cardiovascular remodeling in hypertensive rats,and examine the interference effects of perindopril and spironolactone. Method: The hypertensive model rats were established by constricting the abdominal aorta. 40 male Wistar rats were used in this study and divided into 4 groups at ramdom:① model group,11 rats, hypertensive model rats;② peri group, 11 rats, hypertensive rats subjected to perindopril treatment ,2mg.kg -1 .d -1 ), ③spi group, 11 rats, hypertensive rats subjected to spironolactone treatment(20mg.kg -1 .d -1 ); ④sham group, 9 rats, sham-operated rats serving as control. After 8 weeks the structure and function of the cardiac and aortic were measured by altrosonography; 12 weeks later systolic blood pressure( SBP ) and diastolic blood pressure(DBP)were detected by carotid canula. Results: Compared with sham group,the SBP, LVW/BW ,LAD, thickness of heart and aorta of rats in model group increased significantly(P0.01).Both spi and peri could reversed cardiovascular remodeling and improve left ventricle diastolic dysfunction,moreover spir was obviously better than peri. Conlusion:Spironolactone and perindopril could reverse cardiovascular remodeling and improve heart diastolic function,moreover, spironlactone is better than perindopril.
