Effects of Hygromycin on Induction and Differentiation of Medicago sativa L Callus

Abstract

Hygromycin is a selected marker for genetic manipulation. Effect of hygromycin on callus formation and differentiation of M. sativa leaves was studied. The results were shown that inducement and differentiation of M. sativa leaves was inhibited by hygromycin. When the concentrations of hygromycin was up to 15 mg/ L, callus was not induced. The callus was not differentiated when the concentration of hygromycin was up to 10 mg/ L in induction and differentiation phase. Consequently,We recommended that the concentrations of hygromycin should be 10 mg/ L in induction and differentiation phase .
