Abstract
To study the effects on induction,growth and differentiation of callus(Saussurea involucrate),the hygromycin and Saussurea involucrate leaves were used as stress and explants,respectively.Cultured on MS+ NAA 2.00 mg/L+6-BA 1.00 mg/L media added different concentration(5 mg/L to 50 mg/L) of hygromycin,the result showed that hygromycin of 8 mg/L was the best concentration for screening calli,there were significantly inhibition on callus induction and growth,and the damagement increased with the concerntration rise of hygromycin,the callus was greatly damaged and it's characters were small,browning and dying with the concentration of hygromycin up to 20 mg/L.Meanwhile,the low concentration(0.5 mg/L to 2 mg/L) of hygromycin could enhance the rate of callus differentiation,the averge frequcnce of callus germination was up to 98.33% on the medium of callus induction added 0.5 mg/L hygromycin.
