Abstract
Objective To construct eukaryotic expression plasmid containing the whole genome of HBV. Methods HBV genome was amplified by one-step PCR. The PCR product was digested by Hind Ⅲ and EcoR Ⅰ and cloned into the pcDNA3.1(+) vector. The constructed plasmid was transformed into E.coli DH5α. Result The amplified HBV genome was inserted into pcDNA3.1(+) and the recombinant plasmid was proved by endonuclease digestion and DNA sequencing. Conclusion The recombinant eukaryotic expression vector containing the whole genome of HBV was constructed successfully.
