Molecular mechanism of DDP on proliferation and apoptosis of human cervical cancer cell lines Caski in vivo

Abstract

Objective To study the effects of DDP on proliferation and apoptosis of human cervical cancer cell lines Caski in vivo.Methods Cytotoxicity assay was tested by MTT method;Cell cycle was analyzed by flowcytometry(FCM);Apoptosis landder was evaluated by agarose gel electrophoresis of DNA;The expression of Bcl-2、bax and Caspase3 proteins was detected by Western blot.Results DDP inhibited Caski cell proliferation in a time and dose dependent fashion.DDP degrased the number of cells in G_2/Mphases(P0.01),increased the population of Sphasesat(P0.01)and incresed apoptosis cells number;DDP may down-regulate the expression of Bcl-2,up-regulate the expression of Bax and avtively the Caspase3.Conclusion DDP may inhibit the proliferation of Caski cell lines through effects on cell cycle and apoptosis.that related molecular mechanism of treating Caski cells using DDP were down-regulation of Bcl-2 and up-regulation of Bax.
