The coexpression of angiogenin and glial fibrillary acidic protein in focal cerebral ischemia-reperfusion model in rats

Abstract

Objective To discuss the changes of angiogenin(ANG)expression levels and its coexpression with the glial fibrillary acidic protein(GFAP)in focal cerebral ischemia-reperfusion model in rats.Methods Thirty adult male SD rats were randomly divided into control,ischemia 2 h and reperfusion 1,3,7,and 14 d groups(n=6 in each group).The rat model of focal cerebral ischemia-reperfusion was established with the suture method.The expression levels of ANG and GFAP at different time points after focal cerebral ischemia-reperfusion and their coexpression were detected by the double immunofluorescent labeling method combined with confocal laser scanning microscope.Results The ANG expression levels(ANG fluorescence intensity)after ischemia-reperfusion 1,3 and 7 d in the ischemic brain areas were 56±21,73±22,and 76±21,respectively.The control group was 35±17.GFAP expression levels(GFAP fluorescence intensity)were 94±32,115±35,and 74±31,respectively.The control group was 62±22.As compared with the control group,ANG and GFAP expression in the different time points had significant differences(P0.01).The confocal laser scanning microscope had observed part of coexistence phenomenon of ANG and GFAP.Conclusion Cerebral ischemia may induce upregulation of ANG and GFAP expression in the ischemic brain areas.
