Influence of 5-Aza-2′-deoxycytidine on the Proliferation,Apoptosis and Cycle of Nasopharyngeal Carcinoma Cell Lines

Abstract

Objective To explore the influence of 5-aza-2′-deoxycytidine(5-aza2dC)on the proliferation,apoptosis and cell cycle distribution of four nasopharyngeal carcinoma cell lines-CNE1,CNE2,5-8F and 6-10B. Methods Nasopharyngeal carcinoma cell lines were treated with different concentrations of 5-aza-2dC.Then MTT assay was used to detect the growth curves of cells.Flow cytometry was adopted to discover cell cycle distribution and apoptosis cells. Results Proliferation of CNE1,CNE2,5-8F,6-10B were all inhibited by 5-aza-2dC treatment,while the 50% inhibiting concentration to CNE1 and 6-10B were lower than to CNE2 and 5-8F.The apoptosis cells were dose-dependently increased after 5-aza-2dC treatment in all four cell lines;CNE1 and 6-10B were more sensitive than CNE2 and 5-8F.CNE1,CNE2 and 6-10B blocked in G0/G1 phase while 5-8F blocked in G0/G1 phase and G2/M phase after 5-aza-2dC treatment,CNE1 and 6-10B were more sensitive than CNE2 and 5-8F. Conclusion To nasopharyngeal carcinoma cell lines of CNE1,CNE2,5-8F and 6-10B,5-aza-2dC can inhibit proliferation,promote apoptosis,and block cell cycle,CNE1 and 6-10B were more sensitive than CNE2 and 5-8F.
