Abstract
Lipase(EC3.1.1.3) was immobilized on magnetic chitosan microspheres by using glutaraldehyde as coupling reagent.Different factors influencing the immobilization were investigated,and the optimum conditions were ascertained.Comparative studies of pH and thermal stability between free lipase and immobilized lipase were also conducted;The experimental results showed that the optimal conditions for preparation immobilized lipase were lipase 5.0mg/100mg carrier,immobilization time 5 h,temperature 40℃,pH=8.04,concentration of glutaraldehyde 10%.The maximum binding efficiency reached 90.08%,and the activity of immobilized lipase was 4034.74 U/g·carrier.Therefore,the immobilized lipase holds excellent adaptability in a wider pH region and thermal stability comparable to free lipase.
