Abstract

Objective To improve the method for loading DNA fragment solution into the microinjection pipette during transgenic microinjection.Method Sterile mineral oil was pre-loaded into the unpulled section of the microinjection pipette from its back before DNA fragment solution was sucked into the pipette tip by programmable microinjector IM 300.Result DNA fragment solution was loaded into microinjection pipette successfully and easily.The balance pressure during microinjection process became steady and the withdrawal of DNA solution from the tip-opening was avoided.Conclusion DNA fragment solution can be loaded into the microinjection pipette conveniently and simply by our method where the programmable microinjector is used.And it also ensures that the DNA fragment solution can be injected into pronuclei with every movement of injecting operation,resulting a raise of success-rate in generating the transgenic animals.
