Abstract
Objective To establish a system for a large number of dendritic cells with the antigen-presenting function from Guinea Pig bone marrow.Methods dendritic cells were cultured,induced and amplified in vitro from bone marrow stem cells of Guinea Pig with GM-CSF and IL-4,and identified by morphology and immune phenotype.Results After cultured 7-14 d,cells displayed typical dendritic cells morphology.Detected by immunocytochemistry,the cultured and induced cells were positive in CD14;after mixed cultured with BCG 24h,the cell in apoptosis reached to 18.95%.Conclusion The dendritic cells from bone marrow stem cells of Guinea Pig were successfully cultured in vitro,induced and amplified by combination of lymphocyte separation medium of Guinea Pig and maneuver screening.These dendritic cells were validated and identified through inverted microscope,fluorescence microscope and flow cytometer.
