Screening of differentially expressed genes in gastric carcinoma in North Sichuan area

Abstract

Objective To compare the differential gene expression between gastric carcinoma and normal gastric mucous membrane for screening the differentially expressed genes in gastric carcinoma.Methods The suppression subtractive hybridization(SSH) was used to construct the cDNA subtracted libraries from poorly differentiated gastric adenocarcinoma.RT-PCR was employed to screen the unusual genes from 5 patients at different stages of gastric carcinoma and normal gastric mucous membrane.Results The cDNA libraries from poorly differentiated gastric adenocarcinoma were successfully constructed.Five specificity sequences(NBPF1,FAM48A,RXFP2,ACTB and QDPR) were obtained.RT-PCR showed that RXFP2 mRNA had the differential expression in gastric cancer.Conclusion The screening of gastric cancer specific gene expression of RXFP2 is important to understand the occurrence and development of gastric carcinoma in the etiology of foundation and guide the clinical treatment.
