Effects of Total Saponins of Panax Notoginseng on the Proliferation of Bone Marrow Strowal Cells During Differentiating into Osteoblast in Vitro Mediated by P38MAPK Signal Transduction Pathway

Abstract

Objective:To study the effects of P38MAPK signal transduction pathway on the proliferation of bone marrow strowal cells during differentiating into osteoblast in Vitro.Methods:BMSCs were isolated from femur and tibia of rats and cultured in osteogenic-induced medium(as induced group),osteogenic-induced medium containing PNS 100mg/L(as PNS group) and PNS group medium with SB203580(10uM)(as SB203580 group).The Proliferation of BMSCs were evaluated by MTT assay,the osteogenic differerntiation were determined by alkaline phosphatase(ALP) activity and Alizarin Red S staining.Results:During induction differentiation,the proliferation,ALP activity and Calcium deposit in PNS group were increased compared with induced group and SB203580 group(P0.05~P0.01).ALP activity and Calcium deposit in SB203580 group has no significant difference compared with induced group(P0.05).Conclusion:The proliferation and osteogenic differentiation of BMSCs were promoted by PNS,P38MAPK is the potential signal transduction pathway.
