Abstract
The mature seeds from five varieties of Festuca arundinacea were used as explants to study the effects of steri-(lization) methods and different hormone compositions on callus induction,proliferation and plantlet regeneration.The results showed the rates of both germination and callus induction were prominently higher in sterilization treatment with seed capsule than in that without seed capsule.The callus induction rates of Houndog5 and Yema2 were the highest on the medium with 10.0 mg/L 2,4-D,while that of Xinxiu and Songge was the highest on the medium with 8.0 mg/L 2,4-D and that of Yehuo didn′t change too much(only increasing 10.5%-11.2%) on the media with 4.0-10.0 mg/L 2,4-D.The proliferation medium with 3.0 g/L gelrite as coagulant instead of 7.0 g/L agar could remarkably promote callus growth and its embryonic transformation.The differentiation medium with 2.0 mg/L BA and 0.05 mg/L NAA was better than the others,and the rate of regeneration of Houndog5 on this medium reached 72.8%.
