Effect of Guangxinqi Capsule on Immune Function of Immunosuppressed Mice

Abstract

Objective To investigate the effect of Guangxinqi capsule on the immune function of immunosuppressed mice.Methods Sixty mice were randomly divided into blank control group,model group,positive control group,and high-,medium- and low-dose Guangxinqi groups,10 mice in each group.The mice in the blank control group and model group were given normal saline,in the positive control group were given levamisole(25 mg/kg),in the high-,medium-,and lowdose Guangxinqi groups were given Guangxinqi capsule at doses of 1 800 mg/kg,900 mg/kg,and 450 mg/kg,respectively.The mice were fed by gavage with gastric volume 10 ml/kg for 14 days.In order to establish immunocompromised mice model,the mice in the high-,medium-,and low-dose Guangxinqi groups,model group and positive control group were given intraperitoneal injection with cyclophosphamide of 80 mg/kg on the 10th day,11th day,12th day and 13th day after Guangxinqi administration,while the mice in the blank control group were injected with normal saline.Then the thymus,spleen weights were measured and the indexes were calculated.Carbon clearance method was used to detect monocyte-macrophage phagocytosis,and colorimetric method was used to detect serum hemolysin content for humoral immune function tests.Results The thymus and speen indexes improved significantly in the high-,medium-,and low-dose Guangxinqi groups in contrast with those in the model group(P 0.05).The carbon clearance rate,phagocytic index and serum hemolysin level increased significantly in the high-,medium-,and low-dose Guangxinqi groups(P 0.05).No significant difference was found among the three Guangxinqi groups with respected to the immune function(P 0.05).Conclusion Guangxinqi capsule can improve immune function in the immunosuppressed mice,which does not show a dose-independent relationship in the dose range.
