Abstract
To generate anti-Aβ1-42 antibodies from human phage display antibody library. Methods: Panning from semisynthetic phage antibody library against Aβ1-42 was conducted .to select specific antibodies. The antigen binding activity was determined by ELISA. The phagemid was transformed to E. coll HB2151 in which it was allowed to express soluble scFv proteins. Results: After 4 rounds of panning, 56 positive clones were obtained. Western blot analysis showed that the soluble scFv bound specifically with Aβ1-42, but not with bovine serum albumin. Conclusion: Human anti-Aβ1-42 antibodies can be isolated from phage antibody library without application of immunization. Our results establishes a basis for immunotherapy of Alzheimer' s disease.
