Abstract
Objective To clone human phage antibodies against TRP-1( tyrosinase related protein 1) from a phage antibody library.Methods Panning of the phage antibody library against TRP-1 antigen was conducted to select specific single-chain antibodies. The antigen binding activity and the specificity of the isolated binders were determined with ELISA.Results After 4 rounds of adhension-amplification-elution panning, 8 clones of scFv which could bind specifically to TRP-1 were obtained. The genes of them then were sequenced and analyzed.Conclusion It is feasible to isolate human anti-TRP-1 phage antibodies by-passing immunization using phage antibody library.
