Expression of platelet-derived growth factor B gene mediated by LipofectAMINE in fibroblasts

Abstract

Objective To gain an expression of platelet-derived growth factor B (PDGF-B) gene in fibroblasts with transfection of recombinant plasmid mediated by LipofectAMINE.Methods The recombined eukaryotic expression vector,pcDNA3.1-hPDGF-B,was constructed and transfected into fibroblasts.Positive clones were selected by G418.The expression of PDGF-B gene in the fibroblasts was detected by reverse transcriptase polymerase chain reaction (RT-PCR) and immunohistochemical staining.Results The level of expression of PDGF-B mRNA in the transfected fibroblasts was significantly increased as compared with that in non-transfected fibroblasts.A great many of brown positive pellets were found in the fibroblasts with PDGF-B gene transfection by immunohistochemical staining.Conclusion PDGF-B gene was transfected into fibroblasts mediated by LipofectAMINE and expressed successfully in the fibroblasts,which may contribute to gene therapy after fibroblasts modified by PDGF-B gene.
